摘    要:目的:通过对比实验,找出一条成本低、纯度高的重组人内皮抑素(rhEndostatin)纯化工艺路线。方法:将rhEndostatin包涵体裂解后用缓冲液稀释至不同尿素(Urea)浓度,上分子筛层析;再将含2-巯基乙醇(2-ME)和不含2-ME的样品分别上分子筛层析;将所得蛋白样品电泳,测定活性,比较结果。结果:使用不含Urea和2-ME的缓冲液所得样品纯度、活性、回收率均很好。结论:用不含Urea和不含2-ME的纯化方法是最佳工艺路线。

Abstract:Objective: To find a low-cost and high-purified technology in purifying recombinant endostatin (rhEndostatin) by using comparative experiments. Methods: The lysate of endostatin inclusion body was diluted with buffers to different concentrations (Urea), then it was chromatographied with molecular sieve: samples with or without 2-mercaptoethanol (2-ME) were chromatographied with molecular sieve. All the purified proteins were electrophoresied, their activities were measured and their results were compared. Results: The purity, activity and recovery rate of samples treated with or without Urea and 2-ME were satisfactory. Conclusion: The method of purification of rhEndostatin by using buffers without Urea and 2-ME is the best route.
