地塞米松对支气管哮喘大鼠气道重塑及IL-8的影响

摘    要:目的探讨支气管哮喘大鼠气道重塑与其血清白介素-8(IL-8)表达的关系以及地塞米松的干预作用。方法以卯白蛋白(OVA)致敏并吸入激发制备大鼠哮喘模型。采用计算机图像分析系统及免疫组化技术结合.测定对照组(C)、模型组(N)、地塞米松早期干预组(E)、地塞米松晚期干预组(L)大鼠气道形态学参数及血清IL-8含量。结果E组、L组、N组大鼠出现气道重塑。其中E组气道重塑程度及IL-8含量较轻、N组较重、L组居中,各组问差异有统计学意义(P〈0.05)。结论IL-8与支气管哮喘大鼠气道重塑程度呈正相关;地塞米松可抑制气道重塑。并且可能通过下调IL-8的表达干预气道重塑。

Effects of dexamethasone on airway remodeling and interleukin-8 in asthmatic rats

Abstract:Objective To explore the relationship between airway remodelling and expression of IL-8 and the effect of dexamethasone in asthmatic rats.Methods Asthmatic model was established using ovalbumin(OVA) antigen suspension and ultrasonicatomizing inhalation with ovalbumin(OVA) eight weeks to sensitize.The changes of airway wall morphologic parameter and the relative content of IL-8 in blood serum in the four groups were determined using immunohistochemical technique and a computer assisted image analysis system.Results The various remodeling in airway were observed at all of the rats in E group,L group and N group.The thicknesses of bronchus wall and airway smooth muscle in E group were significantly thinner than those in L group and N group(P < 0.05),and the expression of IL-8 in blood serum in E group was lower than those of L group and N group(P < 0.05).Conclusion The expression of IL-8 in blood serum of asthmatic rats is related with the thickness of bronchus wall and the thickness of airway smooth muscle in asthmatic rats.DXM can influence airway remodelling by down regulating the expression of IL-8.
