Abstract:To explore the association between 2 tag single nucleotide polymorphisms(SNP) in hepatic lipase(HL) gene and serum lipid levels in type 2 diabetes mellitus(T2DM) in Chinese Han nationality.Rs12462668 and rs10426971 of HL gene were analyzed with fluorogenic single nucleotide extension genotype and oligonucleotide microarray hybridization.The serum levels of CHO,TG,HDL-C,LDL-C,Apo A1 and Apo B were detected by automatic biochemical analyzer.The results showed that the frequencies of AA,GG genotype,allele of rs12462668 and haplotype constructed with the two tag SNPs were differ between the two groups.The serum levels of TG and Apo B were higher,HDL-C and Apo A1 were lower in T2DM group than that in control group.The serum lipid results between two groups showed that genotype of the 2 SNPs were associated with disorder metabolism of TG,HDL-C and Apo A1.When serum lipid levels were compared among different genotypes in each group,it showed that HDL-C levels were associated with rs12462668a and TG levels were associated with rs10426971 in T2DM group.The results suggest that two tag SNPs of HL gene are related to T2DM and may influence lipid metabolism.
