摘 要: | 目的探讨尼莫地平对大鼠多发性硬化轴索损伤保护作用的最佳剂量。方法 70只Wistar大鼠随机分为正常组、MS模型对照组和MS模型治疗组,治疗组按给药剂量又分为0.25mg/kg、0.5mg/kg、1mg/kg、2mg/kg、4mg/kg组,分别在治疗前及治疗后14d、28d进行神经丝蛋白(NF)免疫组化分析及髓鞘组化染色分析;每天进行神经功能评分。结果尼莫地平1mgl/kg、2mg/kg、4mg/kg组的轴索损伤明显减轻,与模型对照组及0.25mg/kg、0.5mg/kg组比较差异有显著性(均P<0.01);1mgl/kg、2mg/kg、4mg/kg组之间差异无显著性(均P>0.05)。结论尼莫地平对多发性硬化轴索损伤有保护作用,尼莫地平1mgl/kg是对大鼠多发性硬化轴索损伤保护作用的最佳剂量。

Abstract: | Objective To study the best dose of protective effect of nimodipine on axonal injury in an experimental model of multiple sclerosis.Methods Seventy female Wistar rats,160~200g,were divided randomly into three groups:normal group,MS group and therapetic group(0.25mg/kg,0.5mg/kg,1mgl/kg,2mg/kg and 4mg/kg).The NF immunohistochemistry and myelin histochemistry were performed before therapy,14th day and 28th day after therapy.The neurological deficit was assessed everyday.Results The number of NF positive axons and myelins in the therapetic groups(1mgl/kg,2mg/kg and 4mg/kg)was significantly increased as compared with MS group and the other therapetic groups(0.25mg/kg and 0.5mg/kg)(all P<0.01).There was no statistic difference among high dose of therapetic groups(1mgl/kg,2mg/kg and 4mg/kg)(all P>0.05).Conclusions Nimodipine provide a novel therapy aimed at axonal protection in multiple sclerosis.The best dose of protective effect of nimodipine on axonal injury in an experimental model of multiple sclerosis is 1mgl/kg. |
