| 摘 要： | 目的:研究外源性突变型p27基因(p27mt)对裸鼠皮下肝癌移植瘤血管生长的影响。方法:用人原发性肝癌细胞株SMMC-7721构建人肝癌裸鼠皮下移植瘤模型,将腺病毒介导的突变型p27(Ad-p27mt组)注射瘤体2,1d后观察移植瘤的血管变化,免疫荧光法检测组织中血管内皮细胞生长因子(VEGF)的表达。结果:Ad-p27mt组移植瘤血管单支、纤细,VEGF表达低于对照组和Ad-Lac Z组(P<0.05)。结论:p27mt抑制裸鼠皮下移植瘤滋养血管生成可能通过降低血管中VEGF表达完成。 |

| Abstract: | Objective To study the effects of mutant p27(p27mt) gene on vascular growth of SMMC-7721(HCC) transplated nude mice.Methods The hepatocellular carcinoma model were prepared in nude mice by subcutaneouly transplanting the human hepatocellular carcinoma line SMMC-7721,then adenovirus mediated mutant p27 gene was injected into the transplanted tumor as Ad-p27mt group.The growth of vascular in transplated tumor was observed after 21 d,the expression of VEGF in vascular was detected by immunofluorescence method.Results Compared with Ad-Lac Z group and control group,the tumor vascular were mostly single and thin,and the expression of VEGF was lower in Ad-p27mt group(P<0.05).Conclusion p27mt could inhibit the growth of vascular nourished transplated tumor by decreasing the expression of VEGF. |
