中性粒细胞体外杀伤旋毛虫肌幼虫的实验观察

摘    要:目的探讨感染旋毛虫大鼠中性粒细胞(Neu)在免疫血清协助下对旋毛虫肌幼虫的杀伤作用。方法采用体外CO2培养法,按分组设计在96孔细胞培养板的板孔中加入Wistar雄性大鼠血液中的Neu、旋毛虫肌幼虫、血清(对照血清或经不同条件处理的血清)及含有10%灭活小牛血清的R/MINI-1640培养液,连续培养2d。显微镜下,在12、24、36、48h等时相观察大鼠Neu、免疫血清及补体对旋毛虫肌幼虫的影响。结果无论是感染鼠或正常鼠的单纯Neu或正常血清对旋毛虫肌幼虫均无杀伤作用。在免疫血清及补体参与下,Neu对幼虫的杀伤作用明显增强,同时灭活补体和IgE与仅灭活补体的免疫血清效果相似。结论在体外培养的条件下,感染鼠或正常鼠的单纯Neu或正常血清均无杀伤旋毛虫肌幼虫的作用,感染旋毛虫大鼠的Neu在免疫血清及补体的协助下对旋毛虫肌幼虫具有较强的杀伤作用。免疫血清在灭活IgE前后对Neu杀伤靶虫作用不明显,说明Neu介导的ADCC效应主要依赖的是IgG,并非IgE。

Abstract:Objective To investigate the cytotoxic activity of neutrophils on the muscular stage larvae of Trichinella spiralis. Method Neutrophils from blood of Wistar male rats were incubated with the muscular larvae and immune serum for two days in wells of 96-well tissue culture plates. The effect of immune serum and complement on the viability of muscular larvae was observed at 12, 24, 36 and 48 h after incubation. Result In the absence of immune serum and complement, neutrophil-mediated cytotoxicity was not observed. In the presence of immune serum and complement, a strong cytotoxicity of neutrophils on muscular larvae was observed. Inactivation of serum IgE had no effect on the cytotoxicity of neutrophils on the larvae. Conclusion Immune serum and complement are required for the cytotoxic killing of muscular larvae. IgG appears to be the mediator involved in neutrophil-mediated cytotoxicity on muscular larvae.
