针刺保肝护脑对脑缺血再灌注肝损伤大鼠SODmRNA表达的影响

摘    要:目的:探讨针刺保肝护脑对脑缺血再灌注肝损伤大鼠SODmRNA表达的影响.方法:将40只SD雄性大鼠随机分为假手术组、模型组、针刺对照组、针刺保肝护脑组.线栓法制作缺血再灌注大鼠模型,测试大鼠行为学、血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、SOD活性、肝组织SODmRNA含量.结果:模型组、针刺对照组和针刺保肝护脑组缺血2h(电针治疗前)时神经功能缺损评分无显著差异;经10次电针治疗结束后:与模型组比较,针刺对照组和针刺保肝护脑组神经功能缺损评分有极显著差异(P<0.01);与针刺对照组比较,针刺保肝护脑组神经功能缺损评分变化更为显著(P<0.05).电针治疗前,与假手术组、针刺对照组和针刺保肝护脑组比较,模型组血清ALT、AST水平显著增高(P<0.01),而血清SOD、肝组织SODmRNA明显降低(P<0.01);经10次电针治疗结束后:与模型组比较,针刺对照组和针刺保肝护脑组血清ALT、AST水平明显降低(P<0.01),同时血清SOD、肝组织SODmRNA显著升高(P<0.01);与针刺对照组比较,针刺保肝护脑组血清ALT、AST、SOD和肝组织SODmRNA各项变化更为显著(P<0.05).结论:针刺保肝护脑对脑缺血再灌注大鼠的治疗效果较单纯头穴针刺更为显著,其治疗机制与促进肝脏SODmRNA表达相关.

Abstract:Objective: To investigate the effect of liver/brain-protecting acupuncture on superoxide dismutase(SOD) mRNA expression in rats with liver injury after cerebral ischemia/reperfusion.Methods: Forty male SD rats were randomly divided into sham-operation group,model group,acupuncture control group,and liver/brain-protecting acupuncture group.A rat model of ischemia/reperfusion was established by occlusion.The behavior of rats was assessed,and the serum alanine transaminase(ALT) and aspartate transaminase(AST) levels,SOD activity,and the SOD mRNA content in liver tissue were measured.Results: The three groups showed no significant differences in neurological deficit scores after 2 h of ischemia(before electroacupuncture treatment).After 10 times of electroacupuncture treatment,the acupuncture control group and liver/brain-protecting acupuncture group had significantly different neurological deficit scores compared with the model group(P<0.01),and the liver/brain-protecting acupuncture group showed more significant change in neurological deficit scores compared with the acupuncture control group(P<0.05).Before electroacupuncture treatment,the model group had significantly higher serum ALT and AST levels(P<0.01) and significantly lower serum SOD activity and SOD mRNA content in liver tissue(P<0.01),as compared with the sham-operation group,acupuncture control group,and liver/brain-protecting acupuncture group.After 10 times of electroacupuncture treatment,the acupuncture control group and liver/brain-protecting acupuncture group had significantly decreased serum ALT and AST levels(P<0.01) and significantly increased serum SOD activity and SOD mRNA content in liver tissue(P<0.01),as compared with the model group,and the liver/brain-protecting acupuncture group showed more significant changes in serum ALT and AST levels,serum SOD activity,and SOD mRNA content in liver tissue P<0.05).Conclusion: Liver/brain-protecting acupuncture has better therapeutic efficacy than scalp acupuncture in the rats with cerebral ischemia/reperfusion,as it can promote SOD mRNA expression in liver tissue.
