NAFLD大鼠部分肝切除术后p21的表达变化

目的 探讨非酒精性脂肪肝性肝病(non-alcoholic fatty liver disease,NAFLD)大鼠部分肝切除术后肝再生过程中p21的表达变化及意义.方法 通过高脂饲料喂养建立非酒精性脂肪肝性肝病大鼠模型(F组),同时设立正常饲料组(C组)作为对照,在喂养12周时对F及C组动物分别进行70%部分肝切除术,分别于术后0、1、12、24、36h收集残肝标本,采用RT-PCR及Western blot法分别检测p21的mRNA及蛋白表达变化;并利用免疫组化法检测PCNA (proliferating cell nuclear antigen)的阳性表达率.结果 对高脂喂养12周的中-重度脂肪肝大鼠进行70%部分肝切除术,建立了NAFLD肝再生动物模型.F组p21的mRNA及蛋白表达水平与C组比较差异有统计学意义(P＜0.01),F组明显高于C组;同时PCNA阳性表达率在术后12、24、36h均明显低于C组同时相点(P＜0.01).结论 NAFLD大鼠部分肝切除术后p21的mRNA及蛋白表达水平明显高于正常对照组,同时NAFLD肝再生中细胞增殖能力下降.

Change of p21 after partial hepatectomy in rats with non-alcoholic fatty liver

Objective To investigate the changes of p21 after the partial hepatectomy in rats with non-alcoholic fatty liver disease(NAFLD).Methods The rat models of non-alcoholic fatty liver disease were established by giving a fat-riched diet.The other rats fed with normal diet were taken as normal control at the same stage during the study.These rats were sacrificed at 0,1,12,24,36h respectively after 70% partial hepatectomy at 12 week and the liver tissues were collected.The RT-PCR and Western blot were used to dectect the change of p21.The positive cell percentage of proliferation cell nuclear antigen(PCNA) was measured by immunohistochemical method.Results The liver regenerative model of rats with non-alcoholic fatty liver disease was estabilished successfully.As compared with the control group the expression of p21 in F group was higher than that of C group at both mRNA and protein level.The positive cell percentage of PCNA was lower than that of C group.Conclusion The expression of p21 in rats with NAFLD after partial hepatectomy was higher than that in control group at both mRNA and protein level.At the same time the regenerative capaticity of liver in F group was decreased.
