肿瘤抑制候选基因4及Raf激酶抑制蛋白在不同级别卵巢浆液性腺癌中的表达

目的探讨肿瘤抑制候选基因4(TUSC4)及Raf激酶抑制蛋白(RKIP)在不同级别卵巢浆液性腺癌中的表达情况。方法收集经手术切除并经病理确诊的卵巢浆液性腺癌标本77例,其中卵巢低级别浆液性腺癌25例,卵巢高级别浆液性腺癌52例;另收集经手术切除并经病理确诊的卵巢浆液性囊腺瘤53例及卵巢交界性浆液性囊腺瘤23例;同时选取其他病因手术切除的正常输卵管组织25例及正常卵巢上皮组织25例进行对照。采用免疫组织化学方法对6组标本的TUSC4及RKIP进行检测,并对其阳性表达率进行比较分析。结果 TUSC4在正常卵巢上皮组织、卵巢浆液性囊腺瘤、卵巢交界性浆液性囊腺瘤及卵巢低级别浆液性腺癌、卵巢高级别浆液性腺癌、正常输卵管上皮组织的阳性表达率分别为80.0%、73.6%、60.9%、48.0%、32.7%和32.0%;TUSC4在卵巢低级别浆液性腺癌组织中的阳性表达率显著低于正常卵巢上皮组织(P<0.05);而正常输卵管上皮组织与卵巢高级别浆液性腺癌组织的TUSC4阳性表达率比较差异无统计学意义(P>0.05)。低级别浆液性腺癌组织中TUSC4的阳性表达率低于卵巢浆液性囊腺瘤(χ2=4.91,P<0.05)及正常卵巢上皮组织(χ2=5.56,P<0.05);正常输卵管上皮组织、卵巢高级别浆液性腺癌组织TUSC4的阳性表达率均低于卵巢交界性浆液性囊腺瘤(χ2=4.02、5.22,P<0.05)、卵巢浆液性囊腺瘤(χ2=12.27、17.63,P<0.01)及正常卵巢上皮组织(χ2=11.69、15.14,P<0.01)。RKIP在正常卵巢上皮组织、卵巢浆液性囊腺瘤、卵巢交界性浆液性囊腺瘤、卵巢低级别浆液性腺癌、正常输卵管上皮组织、卵巢高级别浆液性腺癌的阳性表达率分别为100.0%、84.9%、69.6%、64.0%、52.0%和13.5%。卵巢高级别浆液性腺癌组织中RKIP的阳性表达率显著低于正常卵巢上皮组织、卵巢浆液性囊腺瘤、卵巢交界性浆液性囊腺瘤、卵巢低级别浆液性腺癌、正常输卵管上皮组织(χ2=52.06、53.59、23.61、20.59、13.04,P<0.01);卵巢交界性浆液性囊腺瘤组织中RKIP阳性表达率低于正常卵巢上皮组织(χ2=8.91,P<0.01);正常输卵管上皮组织、卵巢低级别浆液性腺癌组织RKIP的阳性表达率均显著低于正常卵巢上皮组织(χ2=15.79、10.98,P<0.01)、卵巢浆液性囊腺瘤(χ2=9.65、4.36,P<0.05)。结论 TUSC4及RKIP均参与了卵巢低级别浆液性腺癌的发病过程,而RKIP亦参与了卵巢高级别浆液性腺癌的发生,且卵巢低级别浆液性腺癌与卵巢高级别浆液性腺癌可能存在不同的分子发病机制。

Expression of tumor suppressor candidate gene 4 and Raf kinase inhibitor protein in different grade of ovarian serous carcinoma

Objective To evaluate the prognosis and guide clinical treatment by studying expression of tumor suppressor candidate gene 4（TUSC4） and Raf kinase inhibitor protein（RKIP） in different grade of ovarian serous carcinoma（OSC）.Methods Seventy-seven cases of OSC specimens which were given resection and diagnosed by pathological analysis were divided into ovarian junior serous carcinoma group（25 cases）,ovarian senior serous carcinoma group（52 cases）;53 cases ovarian serous cystadenoma which were given resection and diagnosed by pathological analysis and 23 cases ovarian borderline serous cystadenoma were also enrolled in the study;25 cases normal oviduct tissue which were given resection for other reasons and 25 cases normal ovarian epithelial tissues were chosen as control. Using immunohistochemical method to detected the expression of TUSC4 and RKIP in the six groups,and the positive expression rates of TUSC4 and RKIP were compared and analyzed in the six groups. Results The positive expression rate of TUSC4 of normal ovarian epithelial tissues,ovarian serous cystadenoma,ovarian borderline serous cystadenoma,ovarian junior serous carcinoma,ovarian senior serous carcinoma and normal oviduct tissues was 80. 0%,73. 6%,60. 9%,48. 0%,32. 7%,32. 0%,respectively. The positive expression rate of TUSC4 in ovarian junior serous carcinoma was lower than that in ovarian serous cystadenoma（χ^2= 4. 91,P〈0. 05） and normal ovarian epithelial tissues（χ^2= 5. 56,P〈0. 05）.
