干眼颌下腺移植术后泪液蛋白分析

摘    要:目的分析自体颌下腺游离移植治疗严重干眼术后分泌的泪液蛋白成分及其与正常泪液的异同。方法收集自体颌下腺游离移植成功后的8例患者的泪液,测定其总蛋白含量及淀粉酶、表皮生长因子(EGF)、分泌性免疫球蛋白A(SIgA)等主要蛋白成分的含量,同时通过不连续的SDS-PAGE蛋白电泳观察其形成的蛋白电泳条带,并与正常人的泪液对比。结果自体颌下腺游离移植后分泌的泪液总蛋白含量明显低于正常泪液(P<0.05),SIgA含量接近正常泪液(P>0.05),EGF含量高于正常泪液(P<0.05),而其所含的淀粉酶浓度则远高于正常泪液(P<0.05)。SDS-PAGE蛋白电泳显示颌下腺移植术后分泌的泪液与正常泪液具有相似的电泳条带及相近的蛋白成分。结论自体颌下腺游离移植术后分泌的泪液具有与正常泪液相近的蛋白成分,但其总蛋白含量较低,淀粉酶含量较高。

Abstract:Objective To analysis the tear protein component of severe xerophthalmia treated with submandibular gland(SMG) transplantation. Methods Tears from 8 patients(8 eyes) with a viable SMG graft(SMG tears),as well as normal tears(20 normal subjects/20 eyes),were sampled.Total protein content,secretory immunoglobulin A(SIgA),amylase,epidermal growth factor(EGF) were measured and SDS-PAGE electrophoresis was performed. Results Compared with normal tears,total protein contnet of SMG tears was lower( P <0.05),SIgA was more similar( P >0.05),but EGF and amylase were higher( P <0.05).SDS-PAGE electrophoresis of SMG tears showed similar protein pattern to normal tears. Conclusions Protein component of tears resulting from SMG transplantation was similar to that of normal tears,but total protein content was lower,EGF and amylase were higher.
