膳食对2型糖尿病大鼠胰岛素抵抗和肝糖原合成酶mRNA表达的影响

摘    要:
目的 探讨饮食因素在大鼠胰岛素抵抗 (IR)和 2型糖尿病 (DM )发生中的作用。方法 高糖、高脂和高糖高脂饮食喂养大鼠 4周后注射小剂量链脲佐菌素建立 2型DM大鼠模型 ,采用胰岛素敏感指数 (ISI)评估IR ,采用逆转录聚合酶链反应检测大鼠肝糖原合成酶mR NA水平。结果 各实验组ISI均降低 ,与对照组比较有显著性差异 (P均 <0 .0 1) ;各实验组大鼠肝糖原合成酶mRNA表达均降低 ,与对照组比较有显著性差别 (P均 <0 .0 5 )。结论 高糖、高脂和高糖高脂饮食均可诱发IR ,并且影响 2型DM大鼠肝糖原合成酶mRNA的表达。

Abstract:
Objective It is to investigate the effect of diet to insulin resistance (IR) and hepatic glycogen synthetase mRNA expression. Methods The rats were respectively fed with high sucrose, high fat and high sucrose high fat diets for four weeks. Then they were induced to non insulin dependant diabetes mellitus (NIDDM) models by injected with low dose of STZ. IR was evaluated with insulin sensitive index (ISI). The level of hepatic glycogen synthetase mRNA were detected with RT PCR. Results ISI in each experiment group was decreased. Compared with that of control group, there was significant difference ( P <0.05). The level of hepatic glycogen synthetase mRNA expression in each experiment group was decreased and significantly lower than that of control group ( P <0.05). Conclusion High sucrose, high fat and high sucrose high fat diets can induce IS and affect the expression of hepatic glycogen synthetase mRNA of NIDDM.
