射干麻黄汤对哮喘豚鼠气道EOS凋亡、IL-5mRNA及IL-10mRNA表达的影响

目的 探讨中药射干麻黄汤对哮喘豚鼠支气管肺泡灌洗液(BALF)中嗜酸性粒细胞(EOS)凋亡、嗜酸性粒细胞阳离子蛋白(ECP)水平、白细胞介素(IL)-5mRNA及IL-10mRNA表达水平的影响.方法 40只健康雄性Hartley系豚鼠随机分为射干麻黄汤组、地塞米松组、哮喘组和正常对照组.采用原位末端标记技术(TUNEL)、荧光酶标记法和RT-PCR技术检测BALF中EOS凋亡、ECP水平、IL-5mRNA及IL-10mRNA表达水平.结果 射干麻黄汤组BALF中的EOS明显较哮喘组低,EOS凋亡率明显较哮喘组高(P＜0.01); ECP水平明显较哮喘组低(P＜0.05);IL-5mRNA水平明显较哮喘组低,IL-10mRNA水平明显较哮喘组高(P＜0.05).结论 射干麻黄汤可以通过减少IL-5mRNA的表达,增加IL-10mRNA的表达有效抑制哮喘豚鼠气道EOS的上升,增加EOS的凋亡;降低ECP水平.

Effects of She Gan Ma Huang soup on apoptosis of eosinophils and the expression of IL-5 and IL-10 mRNA in bronchoalveolar lavage fluid in guinea pigs with asthma

Objective To investigate the effects of She Gan Ma Huang soup on apoptosis of eosinophils(EOS),eosinophil cationic protein(ECP)level and the expression of IL-5 and IL-10 mRNA in bronchoalveolar lavage fluid(BALF)in guinea pigs with asthma.Methods Forty healthy Hartley male guinea pigs were divided into 4 groups by random:She Gan Ma Huang soup group,dexamethasone group,asthma group and normal control group.By using TUNEL,fluorescence labeling and RT-PCR techniques,the apoptosis of EOS,ECP levels and the expression of IL-5 and IL-10 mRNA were measured.Results The EOS in BALF of She Gan Ma Huang soup group were decreased significantly as compared with the asthma group(P<0.01)and tie apoptosis of EOS increased(P<0.01)significantly.The levels of ECP and the expression of IL-5 mRNA were lower and the expression of IL-10 mRNA higher in She Gan Ma Huang soup group than in asthma group(all P<0.05).Conclusion She Gan Ma Huang soup can suppress the increase of EOS,increase the apoptosis of EOS and reduce the ECP levels effectively by down-regulating the expression of IL-5 mRNA and up-regulating the expression of IL-10 mRNA in airway of guinea pigs with asthma.
