NS398诱导人非小细胞肺癌A549细胞凋亡机制的研究

目的:研究NS398诱导人非小细胞肺癌A549细胞凋亡的机制。方法:通过实验细胞培养、MTT试验、细胞形态变化、A549细胞survivin的mRNA表达、A549细胞p-AKT与survivin的蛋白表达来对NS398诱导人非小细胞肺癌A549细胞凋亡的机制进行分析研究。结果:NS398对A549细胞增殖的影响受到浓度的影响,NS398浓度越大,A549细胞增殖抑制率越大;A549细胞在不同浓度的NS398试剂中表现出不同的细胞形态;survivin的mRNA表达随着NS398浓度的增加而减弱,A549细胞p-AKT与survivin的蛋白表达随着NS398浓度的增加而增强。结论:NS398对A549细胞的凋亡具有诱导作用,使肿瘤生长受到抑制,其抗肿瘤机制与AKT磷酸化的抑制、survivin蛋白表达的下调有关。

Study on NS398 induced human non small cell lung cancer A549 cell apoptosis and its mechanism

Objective To study the mechanism of NS398 induced apoptosis of human non small cell lung cancer A549 cell apoptosis.Methods Experimental cell culture,MTT assay,cell morphological changes of A549 cells,survivin mRNA expression,A549 cells p-AKT and survivin protein expression of NS398 induces apoptosis in human non small cell lung cancer A549 cell apoptosis mechanism analysis.Results The effect of NS398 on proliferation of A549 cells by concentration,NS398 concentration,A549 cell proliferation inhibition rate is high;A549 cells showed different morphology of the cells in different concentrations of NS398 reagent;Survivin mRNA expression and decreased with the increase of NS398 concentration,A549 cells and the protein expression of Survivin and p-AKT enhanced with the increase of NS398 concentration.Conclusion Has the function of NS398 on induction of apoptosis in A549 cells,the tumor growth was inhibited,the expression of survivin protein inhibits,antitumor mechanism and AKT phosphorylation and down-regulation.
