连环蛋白P120在肝细胞癌中的表达及其意义

目的检测肝细胞性肝癌(hepatocellular carcinoma,HCC)中连环蛋白P120(P120catenin,P120ctn)的表达,分析其与临床病理特点之间的关系。方法用免疫组化技术检测58例HCC、17例癌旁肝硬化及8例正常肝组织中P120ctn的表达。结果58例HCC组织中有34例P120ctn(58.6%)膜表达弱阳性或缺失,30例(51.7%)胞质表达,异常表达总比例为65.5%(38/58);17例癌旁肝硬化组织中有6例(35.3%)膜表达弱阳性,8例(47.1%)胞质表达,异常表达总比例为47.1%(8/17);8例正常肝组织全部表现为胞膜正常表达,无胞质表达。P120ctn的异常表达与HCC的包膜形成、淋巴浸润、有无卫星灶、肿瘤分化等相关(P<0.05),而与性别、年龄、瘤体积大小、分期等无关(P>0.05)。结论HCC中存在较高比例的P120ctn异常表达,主要表现为胞膜表达缺失伴胞质异常染色,P120ctn可以作为预测HCC转移潜能和预后状况的有价值的指标。

Expression and clinical significance of P120ctn in hepatocellular carcinoma tissues

Objective To investigate in vivo expression of P120ctn in hepatocellular carcinoma (HCC) and analyze its relationship with pathological characteristics. Methods Immunohistochemical method was performed to detect expression of P120ctn in tissues from 58 cases with HCC, 17 with hepatic cirrhosis and eight healthy controls. Results Of 58 cases with HCC, reduced membranous expression of P120ctn was seen in 34 cases (58.6) but cytoplasmic expression in 30 (51.7), with abnormal expression rate of 65.5 (38/58). Of 17 cases with hepatic cirrhosis, six cases (35.3) had decreased expression of P120ctn and eight (47.1) with cytoplasmic expression, with abnormal expression rate of 47.1. There could be seen normal membranous expression of P120ctn rather than cytoplasmic expression in eight healthy controls. Abnormal expression of P120ctn was associated with capsule integrity, nodal metastasis, satellite focus and tumor cell differentiation (P<0.05), but not with gender, age, tumor size and disease stage (P>0.05) . Conclusions In HCC, there exists high rate of abnormal expression of P120ctn, which is mainly reduced membranous expression combined with abnormal cytoplasmic staining. P120ctn can be used as a valuable index for predicting metastasis and prognosis condition of HCC.
