高效液相色谱法测定膦甲酸钠滴眼液含量

目的建立高效液相色谱法测定膦甲酸钠滴眼液的含量。方法采用Diamonsil C18色谱柱（4.6 mm×200 mm,5μm）,流动相：A液-B液-四己基硫酸氢铵-甲醇（700∶300∶0.25∶100）（pH 4.4）（A液：硫酸钠3.22 g,冰醋酸3 mL,44.61 g.L-1焦磷酸钠6 mL,加水至1 000 mL;B液：硫酸钠3.22 g,醋酸钠6.8 g,44.61 g.L-1焦磷酸钠6 mL,加水至1 000 mL）,检测波长：230 nm,流速：1.0 mL.min-1,柱温：室温。结果膦甲酸钠在25.1~803.2μg.mL-1与峰面积呈良好的线性关系（r=0.999 9）,平均回收率100.1%,RSD=0.62%（n=9）。结论该方法简便、快速、准确、重复性好,可作为膦甲酸钠滴眼液的质量控制方法。

Determination of foscarnet sodium ophthalmic solution by HPLC

Objective To establish an HPLC method for the determination of foscarnet sodium ophthalmic solution.Methods The chromatographic condition was Diamonsil C18 column（4.6 mm×200 mm,5 μm）.The mobile phase was consisted of A-B-tetrahexylammonium hydrogen sulphate-methanol=700∶300∶0.25∶100（v/v/m/v,A：dissolve 3.22 g of sodium sulphate decahydrate in water,add 3 mL glacial acetic acid,6 mL 44.61 g·L-1 sodium pyrophosphate solution and dilute to 1 000 mL with water;B： dissolve 3.22 g of sodium sulphate decahydrate in water,add 6.8 g sodium acetate and 6 mL 44.61 g·L-1 sodium pyrophosphate solution and dilute to 1 000 mL with water.The flow rate was 1.0 mL·min-1,and the detection wavelength was 230 nm at the room temperature.Results The linear range of foscarnet sodium was 25.1~803.2 μg·mL-1（r=0.999 9）.The average recovery of foscarnet sodium was 100.1%,RSD%=0.62（n=9）.Conclusion The method is simple,rapid,accurate and reproducible for the determination of foscarnet sodium ophthalmic solution.
