白术挥发油对人类膀胱肿瘤细胞T24生物学行为的影响

摘    要:目的 研究中药制剂白术挥发油(AMO)对人类膀胱癌细胞T24的抑制作用和诱导凋亡作用.方法 将不同质量浓度的AMO作用于人类膀胱癌细胞T24,采用四甲基偶氮唑蓝(MTT)法检测其在体外对T24细胞生长的抑制作用和其有效的作用浓度;流式细胞术(FCM)分析其诱导T24细胞凋亡的机制;用荧光显微镜观察细胞凋亡形态;用透射电镜进一步观察T24细胞的凋亡形态.结果 MTT法测得AMO对T24细胞的IC50值为100 mg/L,质量浓度为50~200 mg/L的AMO具有诱导人类膀胱癌T24细胞凋亡的作用,且随着药物作用时间延长凋亡率逐渐增加.结论 AMO具有直接杀伤人类膀胱癌T24细胞和促进其凋亡的作用.

Abstract:Objective To study the inhibitory effect and apoptosis of atractylodes macrocepha koidz volatile oil on human urinary bladder cancer T24 cell line and its action mechanism, and provide reference to clinical application. Methods Atractylodes macrocepha koidz volatile oil in different concentrations was used in vitro affecting on T24 cell. MTT assay was used to identify its cell growth inhibiting and to gain the effective concentrations. FCM is used to analyze the mechanism of its action on T24 cell. Morphologic changes of T24 cell were observed by fluorescence microscope and transmission electron microscope. Results IC50 value of atractylodes macrocepha koidz volatile oil on T24 cell was 100 mg/L by MTT assay. The apoptotic cells were induced by atractylodes macrocepha koidz volatile oil at the concentration of 50~200 mg/L, and apoptotic rates gradually increased along with its action time. Conclusion Atractylodes macrocepha koidz volatile oil possess the function of killing directly and promoting T24 bladder cell apoptosis.
