骨髓成纤维细胞无血清培养体系的建立及R蛋白对其生长的影响

摘    要:
R蛋白是一种存在于巨噬细胞膜上的受体蛋白,在调节吞噬作用过程中起着重要作用。除巨噬细胞外,骨髓基质细胞也可分泌R蛋白。本文建立了小鼠骨髓成纤维细胞无血清培养体系,并调查了无血清培养条件下R蛋白对小鼠成纤维祖细胞生长的影响。结果发现:多聚赖氨酸能增强成纤维细胞贴壁能力;成纤维细胞生长在无血清培养体系及含血清培养体系中无显著差异(P>0.05);R蛋白对成纤维细胞生长有一定的促进作用(P<0.05)。

Abstract:
Regulatory protein (R-protein),a kind of receptors existing on the macrophage membrane,plays an important role in the regulation of phagocytosis,and is secreted by bone marrow (BM) stromal cells as well as macrophages [1]. In this study,substituting serum with bovine serum albumin, cholesterol, transferrin, insulin and catalase, we established a liquid serum-free culture system for the growth of mouse BM fibroblasts,in which the effect of R - protein on BM fibroblasts of Balb/c mice was investigated. Colony count and 3H -thymidine incorporation assay showed that, by coating petri dishes or plates with poly-L-lysine,one of attachment factors, the proliferation of fibroblasts in serum-free culture was comparable with that in serum - containing culture, and R - protein could augment 3H -thymidine incorporation. These results indicate that attachment factors are crucial for the attachment of fibroblasts, and R -protein plays a role in the proliferation of mouse BM fibroblasts.
