| 摘 要： | 目的为临床骨髓干细胞治疗提供安全、有效的富含间充质干细胞的浓缩骨髓单个核细胞,研究适用于临床治疗的单个核细胞分离技术。方法应用COBE Spectra血液成分单采系统,设置不同参数分离骨髓单个核细胞,在分离前后进行细胞形态学分类、细胞计数检测、表达CD34、CD133和CD271细胞的检测,计算有核细胞回收率、单个核细胞回收率,应用统计方法分析,确定效率较高的单个核细胞采集参数。结果应用Cobe Spectra 6.1的骨髓处理(BMP)程序进行骨髓单个核细胞的采集,采集线路红细胞浓度颜色选择比色卡比容为3%较5%和7%分离的效率均高,差异有统计学意义,循环次数在7—9次范围内较好,MNC百分比较高。单个核细胞数达75.0%,CD34+达1.17%,CD133+0.63%和CD271+0.53%。结论该方法分离骨髓单个核细胞,安全无污染,具有较高的分离效率,且骨髓可以回输,可为临床提供安全可靠的治疗干细胞,值得推广应用。 |
