骨髓单个核细胞分离技术研究及临床应用

摘    要:
目的为临床骨髓干细胞治疗提供安全、有效的富含间充质干细胞的浓缩骨髓单个核细胞,研究适用于临床治疗的单个核细胞分离技术。方法应用COBE Spectra血液成分单采系统,设置不同参数分离骨髓单个核细胞,在分离前后进行细胞形态学分类、细胞计数检测、表达CD34、CD133和CD271细胞的检测,计算有核细胞回收率、单个核细胞回收率,应用统计方法分析,确定效率较高的单个核细胞采集参数。结果应用Cobe Spectra 6.1的骨髓处理(BMP)程序进行骨髓单个核细胞的采集,采集线路红细胞浓度颜色选择比色卡比容为3%较5%和7%分离的效率均高,差异有统计学意义,循环次数在7—9次范围内较好,MNC百分比较高。单个核细胞数达75.0%,CD34+达1.17%,CD133+0.63%和CD271+0.53%。结论该方法分离骨髓单个核细胞,安全无污染,具有较高的分离效率,且骨髓可以回输,可为临床提供安全可靠的治疗干细胞,值得推广应用。

Abstract:
Objective To study the apheresis procedure of collecting mononuclear cells from bone marrow for clinical treatment.Methods Bone marrow mononuclear cells were separated from bone marrow using COBE Spectra apheresis system.Cell morphology,cell counts,positive expression of CD34+,CD133+,CD271+ were detected,and the recovery rate of nucleated cell and mononuclear cell were also calculated.And the main parameters of the procedure with upper efficiency were set via data analysis.Results The efficiency of collecti...
