摘    要:我们成功地建立了应用荆豆素-Ⅰ标记的磁珠(UEA-I-Dynabeads)分离纯化早孕蜕膜组织血管内皮细胞的新技术。荆豆素-Ⅰ型可特异性地与血管内皮细胞膜表面糖蛋白上的岩藻糖基结合,用胶原酶和胰蛋白酶消化蜕膜组织成单细胞悬液,其内皮细胞阳性率在2%左右,经UEA-I-Dynabeads 纯化后,内皮细胞纯度可达90%以上并试培养成功。实验证明此项细胞分离新技术简单、快速且有效,为进一步研究血管内皮细胞的功能与作用打下了良好的基础。

Abstract:We have developed a technique for the isolation of endothelial cells from first trimester decidualtissue using UEA-I coated magnetic polystyrene beads(Dynabeads).UEA-I is an endothelial-specificmarker which reacts with endothelial cell surface glycoprotein via fucose residue.Endothelial cells werereleased from first trimester decidua by the sequential use of collagenase and trypsin.This dissociationmethod yielded 2% endothelial cells which were further purified to>90% homogeneity by rosettingwith UEA-1-Dynabeads,the endothelial cells were released from the Dynabeads by competitive bindingof fucose.This new method is simple and reproducible and lays foundation for research of endothelialcells.
