白细胞介素-17对人牙周膜成纤维细胞表达RANKL、OPG的影响

摘    要:目的 研究白细胞介素17(IL-17)水平对人牙周膜成纤维细胞(HPDLF)中细胞核因子κB受体活化因子配体(RANKL)、骨保护素(OPG)表达的影响,探讨IL-17与正畸牙根吸收的相关性.方法 采用组织块培养法,在体外建立人牙周膜成纤维细胞系.以不同水平(0、5、10、20、40 ng/mL)的IL-17作用HPDLF 24 h.采用RT-PCR和ELISA法,分别检测RANKL和OPG的mRNA、蛋白的表达.结果 (1)在0 ng/mL组中,HPDLF表达RANKL、OPG.(2)0~20 ng/mL各组中,HPDLF中RANKL的mRNA、蛋白表达量与IL-17水平呈正相关性.(3)5~20 ng/mL各组中,OPG的mRNA、蛋白表达量与IL-17水平呈负相关性.(4)0~20 ng/mL组中,RANKL/OPG的mRNA、蛋白的比值与IL-17水平呈正相关性.结论 在无IL-17刺激时,HPDLF可表达RANKL、OPG.IL-17能够促进HPDLF表达RANKL,同时抑制HPDLF表达OPG,上调RANKL/OPG的比值.

Abstract:Objective To study the effects of different concentration of interleukin(IL)-17 on the expression of receptor activator of nuclear factor-κB ligand (RANKL) and osteoprogeterin (OPG) in human periodontal ligament fibroblasts (HPDLF) and explore the relationship between IL-17 and orthodontic root resorption.Methods HPDLF cell line was established through the tissue pieces culture method in vitro.HPDLF were stimulated by IL-17 with five different concentrations(0,5,10,20,40 ng/mL) for 24 h.The expression level of mRNA and protein of RANKL and OPG in HPDLF were detected by RT-PCR and ELISA,respectively.Results HPDLF expressed RANKL and OPG in 0 ng/mL group.The expression amount of mRNA and protein of RANKL in HPDLF was positive correlation with the concentration of IL-17 in 0 to 20 ng/mL group.The expression amount of mRNA and protein of OPG was negative correlation with the concentration of IL-17 in 5 to 20 ng/mL group.The ralative RANKL/OPG ratio of mRNA and protein were positive correlation with the concentration of IL-17 in 0 to 20 ng/mL group.Conclusion HPDLF expresses RANKL and OPG in the absence of IL-17.IL-17 enhances the expression of RANKL and inhibits the expression of OPG in HPDLF.
