高表达LRP16基因对前列腺癌细胞生物学行为的影响

摘    要:目的研究LRP16基因高表达对前列腺癌细胞增殖及侵袭生长的影响。方法利用稳定转染LRP16基因的细胞系,以MTT法测定细胞增殖活性,以平铺Matrigel胶的Transwell培养小室检测细胞的侵袭生长能力。结果高表达LRP16基因对ALVA41细胞(雄激素受体阳性)的增殖没有影响,而对DU145细胞(雄激素受体阴性)则有促进增殖的作用;高表达LRP16基因可抑制ALVA41细胞的侵袭生长,而对DU145细胞的侵袭生长没有影响。结论高表达LRP16基因促进DU145细胞增殖,而抑制ALVA41细胞侵袭生长,可能与除雄激素受体以外的其他机制有关。

Abstract:[Objective] To study the effects of overexpression of LRP16 on the biological behaviours of prostate cancer cells. [Methods] The prostate cancer cells ALVA41, DU145 and their transgenic LRP16 cells were incubated for 24-96 h, and then the vability were determined by MTT. The cellular invasive ability was measured by Boyden chamber(Transwell assays). [Results] Overexpression of LRP16 stimulated DU145 cells prolieration; Overexpression of LRP16 markedly inhibited the invasive growth of ALVA41 cells. [Conclusion] Overexpression of LRP16 facilitated the proliferation of androgen receptor-negative DU145 cells and reduced the invasive ability of androgen receptor-positive ALVA41 cells, the molecular mechanism of which maybe involved the mediation of androgen receptor and other molecular pathway.
