乙酰唑胺对PC12细胞缺氧损伤的保护作用及其机制研究

摘    要:
目的研究乙酰唑胺对缺氧PC12细胞的保护作用及其机制。方法用三气培养箱建立缺氧模型培养PC12细胞,筛选乙酰唑胺作用于缺氧PC12细胞的最佳浓度,观察细胞形态,测定细胞活力、LDH的含量、细胞周期,实时荧光定量PCR法检测HIF-1α及Caspase-3 mRNA的表达。结果与缺氧模型组相比,乙酰唑胺可明显提高缺氧PC12细胞存活率、降低培养基中LDH的含量,降低G1期、提高S期细胞百分比、提高HIF-1αmRNA的表达、降低Caspase-3 mRNA的表达水平。结论乙酰唑胺具有保护PC12细胞缺氧损伤的作用。

Abstract:
Aim To investigate the protective effect of Acetazolamide on the hypoxia cultured PC12 cells.Methods PC12 cells were cultured in three gas incubator and cell hypoxia models were established,optimal concentration of Acetazolamide on hypoxia cultured cells was selected,cells morphology was observed,cell viability,LDH content,cell cycles were measured,Real-time PCR was used to determine HIF-1α and Caspase-3 gene expression.Results Compared with hypoxia group,Acetazolamide could significantly increase the survival rate of hypoxia cultured PC12 cells,decrease LDH levels,increase the percentage of cells in S phase,reduce the percentage of cells in G1 phase,increase HIF-1α gene expression,reduce the gene expression of Caspase-3.Conclusion Acetazolamide has the protective effect on the hypoxia cultured PC12 cells.
