基于响应曲面分析法对归芎药对活血效应相互作用研究

目的应用响应曲面分析法阐明当归-川芎药对活血效应相互作用的范围、性质与程度。方法采用冰水浴与注射盐酸肾上腺素复制大鼠急性血瘀模型,给予不同配比(0∶1、1∶5、2∶5、2∶3、1∶1、3∶2、5∶2、5∶1、1∶0)不同浓度归芎药对后,观察其对血液流变学各指标的影响,采用多指标综合指数法对各指标进行整合,然后运用响应曲面分析法对整合结果进行分析,应用非线性回归的方法确定量效曲线的各个参数,建立三维响应曲面模型,应用Matlab软件构建响应曲面的三维图形。结果归芎药对配比在2∶1到1.3∶1之间,表现出明显的协同作用(协同作用强度为:-0.9);当归剂量从2～3.5 g、川芎剂量从7～9 g这个区域内,表现出拮抗作用(拮抗作用强度为:0.2);而其他比例并未表现出明显的协同或者拮抗作用。结论本研究结果为中医临床合理地应用当归与川芎提供了科学依据;采用响应曲面分析法定量研究中药配伍药物相互作用,为相关研究提供了思路与方法的参考。

Study on interaction of activating blood effects of the herb pair constituted by danggui and chuanxiong by response surface method

Aim To describe the range,properties and extent of the pharmacodynamic interaction of activating blood effects of Gui-Xiong(danggui and chuanxiong) by response surface method.Methods Based on the acute blood stasis rats model,the effects of Gui-Xiong with different proportions(0 ∶ 1,1 ∶ 5,2 ∶ 5,2 ∶ 3,1 ∶ 1,3 ∶ 2,5 ∶ 2,5 ∶ 1,1 ∶ 0) and different concentrations on hemorheology were observed.Then all indexes were integrated to the total activating blood circulation effect value by comprehensive index method.The interaction was analyzed by response surface method.Model parameters were estimated with nonlinear regression.The three-dimensional response surfaces were constructed with Matlab software.Results The proportion from 2 ∶ 1 to 1.3 ∶ 1 of Gui-Xiong had the synergistic action(the value was-0.9);the dose of danggui ranged from 2 to 3.5 g,and chuanxiong from 7 to 9 g showed antagonist action(the value was 0.2),and the other proportions hadn’t the obvious addition and antagonism action.Conclusions The research results can provide scientific evidence for reasonable application of danggui and chuanxiong in traditional Chinese medicine in clinic.Quantitative analysis on drug interactions of herbal compatibility by response surface method can provide reference for relevant studies.
