摘    要:目的 探讨体外培养大鼠骨髓间充质干细胞的可行性,分析其部分表型特点,为细胞移植治疗中枢神经系统疾病奠定基础.方法 采用贴壁筛选法分离纯化大鼠MSCs,进行培养,传代扩增,免疫组化鉴定细胞是否表达具有干细胞特性的标记抗原-神经巢蛋白(nestin),并用流式细胞仪进行荧光检测初步鉴定.结果 分离后的MSCs出现增殖性生长,免疫组化显示巢蛋白(nestin)表达阳性,经流式细胞仪检测显示CD44,CD90阳性,CD45阴性.结论 在体外可培养出较大丰度大鼠骨髓间充质干细胞,而且此方法简单易行.

Abstract:Objective To explore the feasibility of the cultivation of rat marrow mesenchymal stem cells in vitro and analyse their phenotypical properties,to investigate the possibility of a new cell source for central nervous system transplantation.Methods Rat MSCs were isolated and purified from bone marrow,then successive subculture and amplification.nestin,a marking proteins of stem cells were detected by immunohistochemistry,and preliminarily identified by flowcytometer.Results The separated MSCs increase in the medium.Immunohistochemistry staining showed that the MSCs expressed nestin.Flowcytometry showed that it is positive CD44,CD90 negative CD45.Conclusion It is possible to cultivate the high density rat MSCs in vitro.The method is easy to operate and simple.
