痰液MicroRNAs检测在非小细胞肺癌诊断中的应用价值

摘    要:目的检测肺癌患者痰中miRNA-21、miRNA-155、miRNA-137及let-7a的表达,探讨其在肺癌诊断中的价值。方法选取非小细胞肺癌(NSCLC)患者24例。肺良性疾病(OPD)患者24例。11例健康志愿者为对照。Real-time PCR检测痰中miRNA表达。结果 NSCLC患者痰中miRNA-21、miRNA-155表达均显著高于OPD患者及健康对照者,P均<0.01。NSCLC患者let-7a表达均显著低于OPD患者和健康对照者,P均<0.001。三组患者痰中miRNA-137表达无统计学差异,P均>0.05。miRNA-21、miRNA-155及let-7a从OPD中诊断NSCLC的ROC曲线下面积依次为0.863、0.840和0.887,P均<0.01。结论痰miRNAs可能成为肺癌诊断的一种新指标。

Abstract:Objective To detect the expression of miRNA-21, miRNA-155, miRNA-137 and let-7a in sputum of patients with non-small cell lung cancer ( NSCLC), and to investigate their diagnostic significance for lung cancer. Method The study selected 24 pa- tients with NSCLC, 24 patients with other benign pulmonary diseases (OPD), and 11 healthy people as the controls. The expression of miRNA in sputum was detected by Real-time PCR. Results The expression levels of miRNA-21 and miRNA-155 in sputum were signifi- cantly higher in the NSCLC group than in the OPD group and the control group. ( P 〈0.01 ), but the expression of let-7a was just contrary to the above. There was no significantly difference of miRNA-137 expression among the three groups ( P 〉 0. 05 ). Conclusion The measurement of altered miRNAs expression in sputum could be a useful noninvasive approach for the diagnosis of lung cancer.
