穴注黄芪、当归注射液对大鼠胃癌前病变胃粘膜上皮细胞影响的定量分析

目的探讨穴注黄芪、当归注射液对大鼠慢性萎缩性胃炎(CAG)胃粘膜上皮细胞超微结构的影响.方法采用N-甲基-N-硝基亚硝基胍(MNNG)诱发大鼠CAG.随机将70只大鼠分为正常组,模型1组(40μg/mLMNNG造模)及模型2组(60 μg/mL MNNG造模),不作任何治疗.穴注1组(40μg/mL MNNG造模+穴注)与穴注2组(60μg/mL MNNG造模+穴注),造模10周开始以黄芪、当归注射液等份混合注入足三里穴.31周时,进行胃粘膜常规病理及胃粘膜上皮细胞主要细胞器的定量分析.结果1)随造模浓度的增加,胃粘膜损伤及胃粘膜萎缩程度等均相应加重;胃粘膜上皮细胞线粒体退变,粘原颗粒减少,细胞间隙增宽(均P＜0.05).2)穴注法可明显改善胃粘膜上皮细胞功能,缩小上皮细胞间隙,保护胃粘膜屏障而逆转CAG(均P＜0.05).结论胃粘膜上皮细胞的损伤与胃壁屏障结构破坏,胃粘膜萎缩发生、进展至癌变密切相关;穴注法可通过改善上皮细胞的结构与功能,保护胃壁屏障而有效防治CAG.

Quantitative Analysis of Huangqi Injection and Danggui Injection on the Epithelial Cells of Gastric Musosa of Gastric Precancerous Lesion

Objective: To discuss the influenu of Huangqi(Radix Astragali)Injection and Danggui(Radix Angelicae Sinensis)Injection on the epithelial cells of gastric mucosa in rats.Methods: 70 rats were divided into model group 1,model group 2 treatment 1 group,treatment 2 group and normal group.From 9 to 31 weeks Huangqi Injection were injected on Zusanli in treatment 1 group and 2 seperately.Then the gastric pathological test was done,and the main organelles in gastric epithelial cells was analyzed quantitatively.Results: The lesion of gastric mucosa was more serious in model 2 group than in model 1 group.In gastric mucosa,the mitochondrion was degenerated and the intercellular space was enlarged(P<0.05).Acupoint injection in this experiment could treat gastric precancerours lesion by improving the funcion of gastric epithelial cell,shortening the intercellular space and protecting the gastric barrier(P<0.05).Conclusion: The lesions of gastric epithelical cells are associated with the lesion of gastric barrier and gastric precancer.Acupoint injection can prevent the gastric precancerous lesion effectively by improving the structure and function of epithelial cells and protecting the gastric barrier.
