硫酸软骨素A对NIH-3T3鼠成纤维细胞及人皮肤成纤维细胞增殖的影响

摘    要:用MTT法检测了硫酸软骨素A(4-硫酸软骨素,CS-A)对NIH-3T3鼠成纤维细胞和人皮肤成纤维细胞存活与增殖的影响。发现所侧OD值随CS-A浓度的增高而增大,两者呈正相关(3T3鼠成纤维细胞:r=0.8154,P<0.05;人皮肤成纤维细胞:r=0.915,P<0.01),表明CS-A对这两类成纤维细胞的存活和增殖有促进作用。采用PAP和ABC免疫细胞化学染色,观察到两类细胞的胞膜上有CS分布,提示CS可能参与膜结构的组成,对细胞-细胞与细胞-基质之间的相互作用有重要意义;为进一步探讨硫酸软骨素在肝纤维化中的作用提供了可靠的实验依据。

Abstract:By the MTT method we found chondroitin sulfate-A (chondroitin-4-sulfate, CS-A)could promote survival and proliferation of mouse NIH-3T3 fibroblasts and human skin fibroblasts. There was a positive correlation between OD values and CS-A concentrations. The result indicated that fibroblasts could grow well in the condition with high level of CS in fibrotic liver. This CS may accelerate fibroplasia. PAP and ABC stainings showed that CS were present on the surface of both kinds of cells, suggesting that CS may be an integral component of plasma membrane and may take part in cell-cell and cell-matrix interaction.
