摘    要:
目的 探讨低氧后体外培养星形胶质细胞超微结构的变化。方法 通过胶原酶消化、尼龙网过筛、差速粘附等技术对新生小鼠星形胶质细胞进行体外培养,待细胞铺满瓶底时,在培养液表面覆以液体石蜡形成低氧环境,常规培养作为对照,电镜下观察细胞超微结构的变化。结果 正常星形胶质细胞线粒体丰富,可见少量高尔基复合体和粗面内质网。低氧星形胶质细胞表面伸出许多微绒毛样突起,线粒体肿胀,大量溶酶体形成。结论 低氧时星形胶质细胞损伤和修复并存。

Abstract:
Objective To observe the ultrastructural changes of cultured astrocytes in hypoxia. Methods Astrocytes isolated from brains of newborn mice were prepared by collagenase digestion, filtration, differential conglutination. Cells from a confluent cell monolayer were cultured under liquid paraffin to make hypoxic conditions in order to observe the ultrastructural changes(routine culture was made as control). Results Normal astrocytes posses well|developed mitochondria. The Golgi apparatus and rough endoplasmic reticulum are poorly developed. After hypoxia, Microvilli increased and mitochondria swelled, lysosomes are numerous. Conculsion Astrocytes kept both damage and plerosis after hypoxia.;
