摘    要:为了观察IL 2和IL 4对白血病细胞CCR9分布和功能的调节作用 ,我们对 2 1例T ALL患者的血液标本进行分析。流式细胞仪检测发现T ALLCD4 + T细胞高表达CCR9(83 0 %± 7 0 % ) ,IL 2和IL 4联合作用后CCR9的表达显著降低 (16 0 %± 4 0 % ) ;RT PCR、Northernblot和Westernblot及免疫荧光数字共聚焦显微镜检测发现 ,IL 2和IL 4联合作用后T ALLCD4 +T细胞上CCR9的mRNA和蛋白质的表达水平没有改变 ,只是位置发生变化 ,即CCR9发生了内置 ;功能检测发现IL 2和IL 4联合作用亦可相应抑制这些细胞上CCR9的黏附功能和趋化功能。实验结果可能为白血病细胞因子的治疗提供一定的线索

Abstract:To observe the regulatory effects of I L-2 and IL-4 to the expression of the CC chemokine receptor CCR9 on T cell lineag e (T-ALL) CD4+ cells. The blood samples of 21 typical cases of acute lymphocytic leukemia of the T-ALL were analyzed in the prese nt study. It was found that CCR9 was selectively and frequently expressed on the T-ALL CD4+T cells(83.0%±7.0%), and IL-2 and IL-4 in combination could down-regulate the expressions of this receptor on T-ALL CD4+T cells(16.0%±4.0%). However, after the combination action of IL-2 and IL-4, there was no any change on the CCR9 mRNA of T-ALL C D4+T cells and the expression level of proteins, except the internalization of CCR9 on these cells, as demonstrated by RT-PCR, Northern blot and Western blot analysis, digital confocal microscopy examinations and flow cytometry. The com bination action of IL-2 and IL-4 could also inhibit adhesive and chemotactic functions of CCR9 on these cells. This results might provide some hints for the treatment of leukemia wit cytokines.
