酒石酸锑钾对人胃癌细胞的生长抑制作用

目的 :观察酒石酸锑钾在体外对人胃癌SGC - 790 1细胞生长的影响及其作用机制的初步探讨。方法 :采用MTT法检测不同浓度酒石酸锑钾对胃癌SGC - 790 1细胞生长的影响 ;用Hoechst 332 5 8染色后荧光显微镜观察药物作用后细胞的形态变化 ;流式细胞术检测细胞周期和凋亡。结果 :MTT法显示酒石酸锑钾能明显抑制胃癌细胞生长 ,抑制作用呈时间和剂量依赖性 ;酒石酸锑钾作用后 ,Hoechst 332 5 8染色显示细胞核出现明显的凋亡形态学改变 ;流式细胞仪检测到凋亡峰 ,G2 /M期细胞明显增多。结论 :酒石酸锑钾能抑制SGC - 790 1细胞生长 ,有望用于肿瘤的治疗。

Inhibition of human gastric cancer cell proliferation induced by potassium antimonyl tartrate

Objective To study the effects of potassium antimonyl tartrat on human gastric cancer cells(SGC-7901) and to explore its possible mechanism.Methods Human gastric carcinoma cell line SGC-7901 was cultured in vitro. Potassium antimonyl tartrate of different concentrations was added into the media.Cell without adding potassium antimonyl tartrate was used as control. The growth inhibition of cells induced by various concentration in different time course was analyzed by using MTT assay;The cell were stained by Hoechst33258 and the morphologic changes were observed under fluorescence microscope.The apoptosis peak of the cells and the changes of cell cycle were analysed using flow cytomtery.Rusult SGC-7901cells was inhibited obviously by potassium antimonyl tartrate;Chromatin condensation and nuclear fragmentation were seen under fluorescence microscope in the cells stained with Hoechst 33 258.Apoptosis peak were also detected by flow cytometry ,the percentage of G 2/M cells increased significantly in the test group .Conclussion Potassium antimonyl tartrate significantly inhibits the proliferation of SGC-7901 cells.
