摘    要:目的 :研究 1 hydroxy 2 ,3 ,4,6 tetramethoxy xanthone对大鼠局灶性脑缺血再灌注损伤的保护作用。 方法 :采用线栓法阻断大脑中动脉 (MCA)血流 ,造成局灶性脑缺血再灌注模型 (MCAO) ;评价大鼠神经行为功能、测定脑梗塞体积及血清中丙二醛(MDA)含量、超氧化物歧化酶 (SOD)和谷胱甘肽过氧化物酶 (GSH Px)活性。结果 :与大鼠脑缺血再灌注模型组相比 ,1 hydroxy 2 ,3 ,4,6 tetramethoxy xanthone能明显降低脑缺血再灌注大鼠的神经行为学评分分值 (P <0 .0 5 ) ,缩小脑梗塞灶体积 (P <0 .0 1) ,显著提高SOD、GSH Px的活力 (P <0 .0 5 ) ,降低MDA含量 (P <0 .0 5 )。结论 :1 hydroxy 2 ,3 ,4,6 tetramethoxy xanthone对大鼠脑缺血再灌注损伤具有保护作用 ,其作用机制可能与提高SOD、GSH Px的活性 ,减少脂质过氧化有关。

Abstract:Objective:To study the protective effect of 1 hydroxy 2,3,4,6 tetramethoxy xanthone (Xanth) on focal cerebral ischemia reperfusion injurey in rats. Methods: The reversible middle cerebral artery occlusion(MCAO) model in rats was established to investigate the role of Xanth. The scores of neurological deficits and the volume of infarction were detected in MCAO rats. The content of MDA, the activities of SOD and GSH Px was measured in MCAO rats. Results: The scores of neurological deficits and the Volume of infarction were markedly decreased by Xanth in MCAO rats; The content of MDA were obviously decreased while the activities of SOD and GSH Px were significantly increased (P<0.05) by Xanth in MCAO rats. Conclusions: 1 hydroxy 2,3,4,6 tetra Methoxy xanthone could prevent MCAO rats from provides ischemia reperfusion injury , the protective effect is related to it increasing SOD and GSH Px activities and reducing the content of MDA.
