摘    要:目的:建立快速、早期、敏感的诊断人巨细胞病毒(human cytomegalovirus,HCMV)感染的DNA检测方法,旨在了解异基因造血干细胞移植受的HCMV的感染情况及指导临床诊治。方法:利用ELISA和巢式PCR方法对30例异基因造血干细胞移植受进行检测。结果:检测异基因造血干细胞移植受30例,PCR和ELISA方法HCMV阳性分别为20例和18例;其阳性率分别为66.7%和60%。作为阴性对照的20名正常同时进行HCMV的检测,其结果均为阴性。结论:PCR具有早期、快速、简便的优点,适用于临床对HCMV的早期快速诊断和作为指导临床用药及愈后判断的主要手段。

Abstract:Objective To establish a rapid,early and sensitive diagnostic technique for human cytomegalovirus(HCMV) infection after allogeneic hematopoietic stem cell transplantation and discuss its clinical significance.Methods ELISA and nested PCR were used to detect the precent of HCMV in 30 patients of allogeneic hematopoietic stem cell transplantation.Results Among 30 recipients of allogeneic hematopoietic stem cell transplantation ,18 were postive in the ELISA test for HCMV antigen and 20 were HCMV positive based on the nested PCR screening.The positive rate in ELISA and PCR screening the were 66.7% and 73.3% respectively. 20 healthy people (HCMV sem-negative) were also tested with ELISA and PCR.Conclusion Nested PCR is an accurate, rapid and sensetive for early detection of HCMV. It is useful in the early diagnosis of HCMV infection and may be used as the principal method for prognosis of allogenic hematopoietic stem cell transplantation.
