硫胺素对小鼠顺铂肾毒性的防护作用

摘    要:硫胺素(150umol·kg ̄(-1),im)于顺铂(30umol·kg ̄(-1),ip)前1.5h首次给予小鼠,随后每1.5him半量,连续4次,可显著减轻上述剂量顺铂引起的血尿素氮水平升高和近髓肾单位小管的坏死。经硫胺素处理的小鼠,0-24h期间肾及尿内铂含量明显低于单用顺铂组(P<0.01),血浆铂含量无变化。肾内铂含量的减少可能与硫胺素防护顺铂肾毒性有关。

Abstract:The nephrotoxicity induced by cisplatinfcisdiamminedichloroplatinum) at the toxic dose t30 pmol·kg ̄(-1) ip) and charactenzed by high blood urea nitrogen leveland acute tubular necrosis in the juxtamedullary nephron ofmice, was significantly attenuated by pretreatment withthiamine (150 umol · kg ̄(-1), im). 1.5 h prior to the initialcisplatin intection (30 umol · kg ̄(-1), ip), followed by four injec-tions of thiamine (75 umol · kg ̄(-1), im) afler each 1.5 h(P<0.01). Renal and urine platinum contents of mice giventhiamine were significantly lower than those in mice treatedwith cisplatin alone during 0-24 h period post dosing(P<0.01), while the plasma platinum level revealing nochange. These results indicate that the reduction of platinumcontent in the kidney might account for the protection ofthiamine against the renal toxicity.
