槲寄生凝集素蛋白诱导肿瘤细胞凋亡的实验研究

摘    要:目的研究从中药槲寄生中制备的槲寄生凝集素蛋白诱导肿瘤细胞凋亡机制。方法用经典的染色技术:流式细胞术、MTT法分析经分子生物学方法分离纯化的槲寄生凝集素蛋白诱导肿瘤细胞凋亡形态特征,AP峰值和抑制肿瘤细胞增殖的抑制率以研究其诱导肿瘤细胞凋亡机制。结果槲寄生凝集素蛋白具有诱导肿瘤细胞凋亡和显著抑制和阻断肿瘤细胞增殖且以Caspase依赖的方式发挥其生物学作用的。结论槲寄生凝集素蛋白具有直接抗肿瘤作用,在肿瘤免疫学的基础和临床应用研究中具有前景。

Abstract:Objective To investigate the mechanism of the tumor apoptosis induced by a mistletoe lectin protiem(MLP), which was purified from Chinese mistletoe. Methods ML was purified by molecular biological methods, classical dyeing and flow cytometry. MTF was used to analyze the morphological features, AP peaks, proliferation inhibition rates of the tumor apoptosis cell induced by MLP. Results MLP was found that ( 1 ) induced the apoptosis of tumor cells; (2) inhibited and blocked tumor cell proliferations (3) played its biological role through the caspase-3 dependent pathway. Conclusion MLP has a direct antitumor effect, and possesses good prospects in the basic and clinical application research of the tumor immunology.
