蚓激酶导向溶栓免疫纳米粒的制备工艺研究

摘    要: 目的制备具有导向溶栓作用的蚓激酶纳米粒。方法利用溶剂挥发法制备白蛋白纳米粒,与单抗相接制备了蚓激酶免疫纳米粒;对纳米粒的载药量、大小及分布、形态及体外释放等进行了研究。结果纳米粒平均粒径为68nm,跨距为91nm,包封率为(86.62±1.65)%,载药量为(3.96±0.63)%;电镜结果显示,外形圆整,呈球形,分散良好,相互之间不粘连;其体外释药实验表明明显的缓释性能。结论该导向溶栓纳米粒制备工艺可行。

Abstract:OBJECTIVE To prepare lumbrokinase immunonanoparticles of thrombolytic. METHODS The lumbmkinase nanoparticles were prepared by solvent-evaporation proeess, and lumbrokinase immunonanoparticles were prepared by linked nanoparticles with monoclonal antibody. The drug loading, entrapment efficiency, diameter of nanoparticles and particle diameter distribution, shape and release curve of nanoparticles were investigated in vitro. RESULTS The average diameter of lumbrokinase nanoparticles was 0.068 μm, the span was 0.091 μm, the entrapment efficiency was (86.62+1.65)% and drug loading was (3.96+0.63)%. The shape of nanoparticles was round without conglomeration. The study showed that the lumbrokinase immunonanoparticles have a obvious sustained release effect. CONCLUSION The preparation of lumbrokinase immunonanoparticles of thrombclytic is feasible.
