摘    要:目的:研究三氧化二砷(As2O3)对胃癌细胞株SGC7901的生长及血管内皮生长因子(VEGF)表达的影响.方法:体外培养胃癌细胞株SGC7901,加入不同浓度的As2O3干预,用Western bloting和荧光定量RT-PCR法测定VEGF蛋白和mRNA表达,加入外源性重组人VEGF165,观察VEGF对肿瘤细胞生长的刺激以及As2O3对此作用的抑制.结果:胃癌细胞株SGC7901经As2O3作用后,VEGF的蛋白表达明显减少,但VEGFmRNA拷贝数无差异.在SGC7901细胞培养中加入外源性VEGF165,其细胞活力有所增加,细胞存活率均大于100%;当同时加入VEGF165和As2O3时,其细胞存活率明显减低.结论:As2O3在蛋白水平抑制胃癌细胞株SGC7901的VEGF表达,抑制SGC7901的生长.

Abstract:Objective:To investigate the effect of As2O3 on the expression of vascular endothelial growth factor (VEGF) in gastric cancer cell strain SGC7901.Methods:Gastric cancer cells SGC7901 cultured in vitro were interfered with As2O3 at different concentrations.Both protein and mRNA expressions of VEGF were detected by Western bloting and fluorescent quantitative RT-PCR.The stimulation on the cells growth by VEFG and the inhibition of such stimulating effect by As2 O3 were observed,after the addition of external recombinant human VEGF165.Results:With the effect of As2O3 on gastric cancer cell strain SGC7901 VEGF protein expression was obviously reduced,but no significant change occurred in the copies of VEGF mRNAs.The addition of external VEGF165 somewhat enhanced the viability of SGC7901 cells,resulting in more than 100% of survival rate for all the cells,simultaneous addition of VEGF165 and As2O3,however,caused remarkably reduced cell survival rate.Conclusion:As2O3 can inhibit VEGF protein expression in gastric cancer cell strain SGC7901 as well as the growth of SGC7901 cells,and also has an inhibiting effect on tumor growth and angiogenesis by preventing VEGF autocrine and paracrine pathways.
