Abstract:
Objective To observe brain-derived neurotrophic factor(BDNF) protein and mRNA levels in the hippocampus of post-stroke depression(PSD) model and the intervention of sertraline.To initial disscuss the role of BDNF in the course of PSD.Methods The focal cerebral ischemia model was set up by the method of middle cerebral artery blocking(MCAO).The rats were suffered Chronic unpredictable mild stress(CUMS)and were separated living to induce the PSD model.Some rats were intervened by sertraline.Application detection of stress 18,28 d,respectively,when the hippocampal expression of BDNF protein and LmBNA levels were detected by western blot and Real time-PCR in 18d and 28d post stess.Results On the 14th day after CUMS,the PSD group showed significantly less locomotor activity and sugar-water consumption(P<0.05 or P<0.01)compared with the control group.On the l 8th,28th day,BDNF protein levels ofthe PSD group declined significantly(P<0.05 or P<0.01)compared with the control group.BDNF mRNA levels on the l8th day were reduced,but there was no statistic difference.When the 28th day,BDNF mRNA levels significantly declined(P<0.01).On the 14th and l8th day,open-field activities and the amount of sugar-water consumption of the fluoxertin intervention group increased markedly(P<0.05 or P<0.01)compared with the PSD group,while onthe 18th and 28th day,BDNF protein and mRNA levels of the sertraline group were significantly increased(P<0.01)compared with the PSD group respectively.Conclusion At the same time of Neurological deficit in PSD rats,the core symptoms of depression were anhedonia and exploratory behavior reduce.And the growth rate of body weight was significantly slowed down.PSD hippocampal BDNF protein and mRNA expression was significantly decreased.BDNF expression increased after the intervention of sertraline.BDNF prompted the initial post-stroke depression in the role.BDNF may has neuroprotect effect in poststroke depression.
