B类1型清道夫受体基因多态性与2型糖尿病下肢血管病变的相关性

摘    要:目的探讨B类1型清道夫受体(SR-B1)基因多态性与2型糖尿病(T2DM)下肢血管病变间的关系。方法选取T2DM患者463例,根据血管超声检查分为合并下肢血管病变和不合并下肢血管病变两组,另选取社区健康体检正常的200人作为正常对照组,抽取受试者静脉血,提取DNA,进行PCR,对PCR产物测序。结果 T2DM合并下肢血管病变、T2DM不合并下肢血管病变及正常对照组间SR-B1外显子8基因型和等位基因频率的差异均无统计学意义(均P〉0.05)。进一步性别亚组分析,3组男性受试者之间SR-B1外显子8基因型及等位基因的差异均有高度统计学意义(均P〈0.01),女性受试者间的差异均无统计学意义(均P〉0.05)。结论 SR-B1外显子8基因多态性与男性T2DM下肢血管病变相关。

Abstract:Objective To investigate the polymorphism of the scavengen receptor class B type 1(SR-B1) and it relationship with type 2 diabetes mellitus(T2DM) complicated with vascular disease of low extremities.Methods Four hundreds and sixty-three hospitalized T2DM patients were divided into vascular disease of low extremities group and non-complications group by the ultrasound test of the lower extremities.Two hundreds people for health care were non-randomly recruited as the healthy controls.The polymorphism of the human SR-B1 gene's intron 5 and exon 8 in 463 T2DM patients and 200 health controls was evaluated by gene sequencing.Results The SR-B1 exon 8 had polymorphism.There was no significantly difference in frequency of genotype between the vascular disease group(all P0.05) of low extremities,non-complications group and the control group.In subgroup analysis,polymorphisms in exon 8 was in strong linkage disequilibrium in men(all P0.01),but not in women(all P0.05).Conclusion In men,SR-B1 polymorphisms show associations with vascular disease of low extremities in type 2 diabetes mellitus.
