MMP-9、TIMP-1在COPD大鼠肺组织中的表达及虎杖的干预影响

摘    要:摘要:目的:探讨虎杖对慢阻肺大鼠模型肺组织内基质金属蛋白酶-9(MMP-9)及金属蛋白酶抑制剂-1(TIMP-1)表达的影响。方法:选用熏烟和气管内注入脂多糖(LPS)的造模方法建立大鼠慢性阻塞性肺病(COPD)模型。40只雄性SD大鼠被随机分成4组,每组10只:正常对照组、COPD模型组、强的松组、虎杖干预组。在每次熏烟前0.5h给药,熏烟的第29天采用虎杖煎剂、强的松进行相关干预,给药28天。在给药第28天,处死大鼠。Real-Time PCR方法检测肺组织匀浆标本MMP-9和TIMP-1表达。结果:与对照组相比,其他3组大鼠MMP-9和TIMP-1表达水平均明显升高(P<0.05);与COPD模型组相比,强的松组、虎杖组表达水平均显著降低(P<0.05)。结论:虎杖可降低MMP-9和TIMP-1表达,对COPD大鼠肺组织有一定保护作用,这也可能是其控制气道炎症及抑制重塑的作用有关。

Expression of MMP-9 and TIMP-1 in Lung Tissue of COPD Rats and the Intervention Effect of Polygonum Cuspidatum
Abstract:Objective: To investigate the effects of Polygoni cuspidata on the expression of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in lung tissue of COPD rats. Methods: Chronic obstructive pulmonary disease (COPD) model was established by fumigation and intratracheal injection of lipopolysaccharide (LPS). Forty male SD rats were randomly divided into 4 groups with 10 rats in each group: normal control group, COPD model group, prednisone group and Polygonum cuspidatum intervention group. 0.5h before each fumigation, the first 29 days of fumigation with Polygonum cuspidatum decoction, prednisone-related intervention for 28 days. On the 28th day of administration, the rats were sacrificed. Real-Time PCR method was used to detect the expression of MMP-9 and TIMP-1 in lung homogenates. Results: Compared with the control group, the expression levels of MMP-9 and TIMP-1 in the other three groups were significantly increased (P <0.05). Compared with the COPD model group, the expression levels of both the prednisone group and the Polygonum cuspidatum group were significant Decreased (P <0.05). CONCLUSION: Polygonum cuspidatum can decrease the expression of MMP-9 and TIMP-1 and protect the lung tissue of COPD rats, which may be related to the control of airway inflammation and inhibition of remodeling.
