Survivin、Bcl-2和Bax在喉鳞状细胞癌中的表达

摘    要:目的探讨凋亡抑制基因survivin在喉鳞癌中的表达及其与Bcl-2、Bax基因表达的相关性。方法应用免疫组织化学链酶卵白素-生物素复合体(strept avidin-biotin complex,SABC)方法检测45例喉鳞癌及10例正常喉黏膜组织中survivin,Bcl-2及Bax基因的表达。结果喉鳞癌组织中,Survivin的表达率为80%,显著高于正常喉黏膜组织的表达率20%(P均<0.05)。Survivin蛋白表达与喉鳞癌的TNM分期及淋巴结转移有关(P均<0.05)。Survivin表达与Bcl-2蛋白表达呈正相关(P<0.05),而与Bax蛋白表达呈负相关(P<0.05)。结论Survivin在喉鳞癌的发生发展中起重要作用,可能成为喉鳞癌基因治疗的新靶点;survivin基因可能与凋亡相关基因Bcl-2、Bax在喉鳞癌的发展中分别起协同和拮抗作用。

Abstract:Objective:To study the expression of survivin and its relationship with the expressions of both Bcl-2 and Bax in locally advanced laryngeal squamous cell carcinoma (LSCC). Methods:Immunohistochemical strept avidin-biotin complex (SABC) method was used to examine the expression of survivin, Bcl-2,and Bax in 10 normal laryngeal mucosa tissues and 45 LSCC tissues. Results:Survivin was expressed in 36 of 45(80%) cases of LSCC and in 2 of 10(20%) cases of normal laryngeal mucosa tissues. Expression of survivin was significantly higher in LSCC than in normal tissues. Over-expression of survivin was related to the tumor grade and cervical lymph node metastasis(P<0.05). There was a positive correlation between expression of survivin and Bcl-2;whereas there was a negative correlation between the expression of survivin and Bax. Conclusion:Survivin may play an important role in the initiation and progression of LSCC. It may be a new therapeutic target in cancer therapy. Survivin may have synergistic or antagonistic effects with Bcl-2 or Bax in the development of LSCC, respectively.
