肾精亏虚对雄性大鼠骨质的影响*

摘    要:目的:探讨肾精亏虚对雄性大鼠骨质的影响。方法:采用房劳的方法建立肾精亏虚模型雄性大鼠为模型组,另设正常组和自然衰老组,比较3 组血清1 型前胶原氨基端延长肽( P1NP)和1 型胶原C末端肽( CTX-1)含量水平,X射线测量全身骨密度,H-E 染色观察胫骨骺端骨小梁分布情况,免疫荧光化学法检测p16 蛋白的表达。结果:3 组P1NP 水平差异无统计学意义;模型组和自然衰老组CTX-1 水平显著高于正常组,模型组与自然衰老组CTX-1 差异无统计学意义;模型组和自然衰老组的骨密度低于正常组,骨小梁稀疏断裂较正常组明显;模型组和自然衰老组骨组织p16 蛋白表达高于正常组,模型组与自然衰老组的p16 蛋白差异无统计学意义。结论:肾精亏虚可促进雄性大鼠骨质疏松。

Effects of kidney essence deficiency on bone mass in male rats*
Abstract:Objective To explore the effect of kidney essence deficiency on bone quality of male rats. Methods Themodel of kidney essence deficiency was established via "Fang Lao" method as a model group, besides, the normalgroup and the natural aging group were established. There were three groups, 10 animals in each group. The serumN. terminal propeptide of type I collagen( P1NP) and cross linked C-telopeptide of type I collagen( CTX-1) levelsof the three groups were compared. Then, general bone mineral density( BMD) was measured by X ray. Finally,the distribution of trabeculae in tibial epiphyseal extremity and the expression of p16 protein were observed by H-Estaining and immunofluorescence, respectively. Results There was no statistical significance in the P1NP levelof the three groups. The CTX-1 level in model group and natural aging group was significantly higher than that innormal group. There was no significant difference between model group and natural aging group. Moreover, the bonedensity of model group and natural aging group was lower than that of normal group, and the sparse fracture of bonetrabecular was more obvious than that of normal group. The expression of p16 protein in model group and naturalaging group was higher than that in the normal group. There was no significant difference between the model groupand the natural aging group. Conclusion Kidney essence deficiency can contribute to osteoporosis of ma le rats.
