高效液相色谱—示差折光检测法测定注射用转化糖中果糖与葡萄糖的含量

摘    要:目的建立同时测定注射用转化糖中果糖与葡萄糖含量的高效液相色谱—示差折光检测法。方法采用氨基键合硅胶柱(250 mm×4.6 mm,5μm),流动相为乙腈-水(85∶15),流速1.00 mL/min,柱温30℃,外标法计算。结果线性范围分别是:果糖0.1266~0.8861 mg(r=0.9995,n=7);葡萄糖0.1263~0.8842 mg(r=0.9999,n=7)。回收率:果糖为99.8%(RSD为0.68%,n=6);葡萄糖为99.7%(RSD为0.69%,n=6)。结论此法分离效果好,快速、简便,适用于该制剂两组分含量的同时测定。

Abstract:Purpose To establish a method for the determination of two constituents fructose and glucose in invert sugar for injection by HPLC-RID.MethodsNH2 column(250 mm×4.6 mm,5 μm)was used.The mobile phase was composed of acetonitrile and water(85∶15),at a flow rate of 1.0 mL/min.The temperature of the column was 30℃.ResultsThe linear response range of fructose was 0.126 6-0.886 1 mg(r=0.9995,n=7)and that of glucose was 0.126 3-0.884 2 mg(r=0.999 9,n=7).The mean recovey of fructose was 99.8%(RSD=0.68%,n=6)and that of glucose was 99.7%(RSD=0.68%,n=6).ConclusionThe method was simple and quick with a good result of determination.This method was suitable for the determination of two constituents in compound preparation at the same time.
