PTEN蛋白在卵巢癌发生发展过程中的表达及临床病理意义

目的:检测抑癌基因PTEN蛋白表达在卵巢癌发生发展中的作用及相关病理机制.方法:采用免疫组化法检测95例卵巢癌组织、19例卵巢交界性肿瘤、10例正常卵巢、10例卵巢囊肿组织中PTEN蛋白表达,比较其表达与卵巢癌临床病理特征的关系.结果:卵巢交界性肿瘤和卵巢癌组织中PTEN蛋白表达显著低于正常卵巢和卵巢囊肿(P＜0.05);PTEN蛋白表达与卵巢癌组织分化程度呈正相关(P＜0.01),与临床病理分期负相关(P＜0.05);卵巢癌中PTEN蛋白表达降低或失活与卵巢癌淋巴结转移相关(P＜0.01);上皮性卵巢癌中子宫内膜样癌PTEN蛋白表达最低(P＜0.01).结论:在卵巢癌发生过程中PTEN蛋白表达降低起到重要作用,表达异常的PTEN蛋白与卵巢癌的组织分化程度、临床病理分期和转移能力相关,与上皮性卵巢癌中子宫内膜样癌PTEN蛋白表达关系更为密切.PTEN蛋白表达可作为判定卵巢癌发生及淋巴转移能力的一项有效客观指标.

Expression of PTEN protein and clinicopathological significance in tumorigenesis and progression of ovarian cancer

Objective: To investigate the role of PTEN protein expression in tumorigenesis and progression of ovarian cancer and its pathological mechanisms. Methods:We examined expression of PTEN protein in 10 cases of normal ovarian tissue, 10 of ovarian cyst tissue, 19 of ovarian borderline tumor, 95 of ovarian cancer by using the immunohistochemistry. The expression was compared with clinicopathological features of ovarian cancer. Results:Expression of PTEN protein was significantly lower in ovarian borderline tumor or ovarian cancer than that in normal ovary or ovarian cyst(P<0.05). Expression of PTEN was negatively correlated with clinicopathological staging of ovarian cancer(P<0.05),but positively correlated with histological differentiation(P<0.01). Loss or reduced expression of PTEN protein was related with lymph node metastasis of ovarian cancer (P<0.01). Among ovarian epithelial cancers, endometrioid cancer expressed was the lowest level of PTEN protein(P<0.01). Conclusion:Reduced expression of PTEN protein plays an important role in tumorigenesis of ovarian cancer. Its altered expression was associated with histological differentiation, clinicopathological staging, and lymph node metastasis of ovarian cancer. Expression of PTEN protein was closely associated with ovarian endometrioid cancers. Decreased expression of PTEN gene might be a useful marker for predicting the carcinogenesis and progression of ovarian cancer.
