高效毛细管电泳法测定萹蓄中金属阳离子的含量

摘    要:目的:建立测定萹蓄中金属阳离子含量的方法。方法:采用高效毛细管电泳法。色谱柱为未涂层弹性融硅石英毛细管柱(50cm×50μm ID,有效长度46cm),运行缓冲液为7.0mmol·L-1咪唑+6.0mmol·L-1酒石酸(pH6.0),分离电压为20kV,电动进样时间为3s(电压为15kV),检测波长为214nm。结果:K+、Na+、Mg2+、Mn2+、Ca2+、Zn2+检测浓度分别在0.5~120.0mg·L-1(r=0.9993)、0.5~120.0mg·L-1(r=0.9987)、0.1~60.0mg·L-1(r=0.9973)、0.1~120.0mg·L-1(r=0.9992)、0.1~60.0mg·L-1(r=0.9991)和0.5~120.0mg·L-1(r=0.9982)范围内与各自峰面积积分值呈良好线性关系;K+、Na+、Mg2+、Ca2+、Zn2+的平均加样回收率分别为103.5%、96.0%、93.4%、105.4%和101.6%,RSD分别为1.75%、1.66%、1.52%、1.43%和1.19%(n=6)。结论:本方法简便、快速、准确、可靠,可用于萹蓄中金属阳离子的含量测定。

Abstract:OBJECTIVE: To establish a method for the determination of metal cations in Polygonum aviculare. METHODS: High performance capillary electrophoresis was adopted. The separation was performed on a uncoated fused silica capillary (50cm×50μm ID, effective length of 46 cm) with 7.0 mmol·L^-1 imidazole+6.0 mmol·L^-1 tartaric acid as buffer solution (pH=6.0). The separation voltage was 20 kV and the sample was injected for 3 s at 15 kV. The detection wavelength was set at 214 nm. RESULTS: The linear range was 0.5-120.0 mg· L^-1(r=0.999 3) for K+, 0.5-120.0mg· L^-1(r=0.998 7) for Na^+, 0.1-60.0 mg· L^-1 (r=0.997 3) for Mg^2+,0.1-120.0 mg· L^-1(r=0.999 2) for Mn^2+,0.1-60.0 mg· L^-1(r=0.999 1) for Ca^2+ and 0.5-120.0mg· L^-1 (r=0.998 2) for Zn^2+. Mean recovery rates of K^+,Na^+,Mg^2+,Ca^2+,Zn^2+ were 103.5% (RSD= 1.75%, n=6), 96.0% (RSD= 1.66%, n=6), 93.4% (RSD= 1.52% ,n=6), 105.4% (RSD= 1.43%, n=6) and 101.6% (RSD= 1.19% ,n=6). CONCLUSION: The method is convenient, rapid, accurate and reliable for the content determination of metal cations in P aviculare.
