供体可溶性组织相容性抗原的制备及其诱导淋巴细胞凋亡的研究

目的:通过静脉注入供体可溶性组织相容性抗原,诱导受体淋巴细胞凋亡。方法:应用3mol/L氯化钾的抽提供体(Wistar大鼠)可溶性抗原,然后将其供体可溶性抗原经静脉注入受体(SD大鼠),采用TUNEL法标记受体胸腺及脾脏淋巴细胞以观察其凋亡情况,受体淋巴细胞对供体抗原的反应采用混合淋巴细胞培养。结果:在体外,制备的供体可溶性抗原可刺激致敏后的受体淋巴细胞增生;在体内,供体可溶性抗原可导致受体胸腺及脾脏的淋巴细胞凋亡。结论:制备的供体可溶性抗原具有免疫学活性,能在体内诱导淋巴细胞凋亡。

Preparation of Soluble Donor MHC Antigen and Its Effect in Inducting Recipient Lymphocytes Apoptosis.

Objective: To induce recipient lymphocytes apoptosis by administration of soluble donor MHC anti-gen. Methods: The donor (Wistar rat) MHC antigen was extracted by means of 3 mol/L KCl. The recipi-ents were then treated by intravenous injection of the soluble donor MHC antigen extracted. The apopto-tic recipient spleen and thymus lymphocytes were labelled with TUNEL. The responsiveness of the recipi-ent lymphocytes to donor MHC challenge was measured by MLC. Results: In vitro, the prepared soluble MHC antigen of the donor stimulated the proliferation of donor lymphocytes. In vivo, apoptosis of the re-cipient spleen and thymus lymphocytes was observed. Conclusions: The soluble donor MHC antigen pos-sesses immunologic activity which could induce apoptosis of specific recipient lymphocytes.
