NF-κB与Survivin基因在脑胶质瘤中的表达

摘    要:目的探讨细胞核因子KB和Survivin基因在胶质瘤中的表达及其与肿瘤细胞凋亡的关系。方法分别采用原位杂交和免疫组织化学S—P法检测80例星形细胞瘤患者和10例正常脑组织Survivin mRNA和NF—KB的表达,用末端转移标记法(TUNEL)检测肿瘤组织的凋亡并计算其凋亡指数(AI)。结果星形细胞瘤组织中Survivin mRNA的阳性表达率明显高于对照组(P〈0.01),高度恶性组的阳性率明显高于低度恶性组(P〈0.01);AI在星形细胞瘤中明显高于对照组(P〈0.01),而Survivin mRNA阳性组低于阴性组(P〈0.01);NF—kB在星形细胞瘤中的表达明显高于对照组(P〈0.01),且Survivin mRNA阳性组明显高于阴性组(P〈0.05)。结论NF-kB与Survivin在星形细胞瘤中的表达密切相关,NF-kB的过表达可能通过上调Survivin mRNA的表达抑制星形细胞瘤细胞的凋亡.从而在肿瘤的发生、发展和治疗中发挥重要作用。

Abstract:Objective To investigate the expressions of nuclear factor KB (NF-kB) and Survivin in astrocytomas, as well as the correlation between them and tumor cells apoptosis. Methods The expressions of Survivin mRNA and NF-kB were detected respectively by in situ hybridization and immunohistochmical (S-P) method in 80 cases of astrocytomas and 10 cases of normal brain tissues. Apoptosis cells were detected by TUNEL and the apoptosis index (AI) was calculated. Results The positive expression rate of Survivin mRNA in the astrocytomas was significantly higher than that in the control group (P〈0.01), and it was significantly hiher in the high grade astrocytomas than that in the low grade ones (P〈0.01). AI in the astrocytomas was significantly higher than that in the control group (P〈O.01) and AI in Survivin mRNA positive group was significantly lower than that in its negative group (P〈0.01). Expression of NF-kB in the astrocytomas was significantly higher than that in the control group (P〈0.01). The expression rate of NF-kB in Survivin mRNA positive group was significantly higher than that in its negative group (P〈0.05). Conclusions There is a close correlation between the empressions of NF-KB and Survivin in the astrocytomas. Overexpressio, of NF-KB probably upregulates the Survivin mRNA expression in the astrocytomas, inhibits tumor cell apoptosis and play an important role in the tumor pathogenesis and development.
