双丹脂质体凝胶抑制兔耳瘢痕增生的作用及机制研究

摘    要:
目的 考察双丹脂质体凝胶抑制兔耳瘢痕增生的作用及可能机制.方法 制备兔耳增生性瘢痕模型,21 d后给药,持续42 d.采集兔耳缘静脉血,分离血清进行IL-1β、TGF-β1含量测定,采集瘢痕组织标本进行HE染色后光镜下观察病理组织学变化,同时测量瘢痕增生指数和成纤维细胞密度.结果 与模型组相比,双丹脂质体凝胶组瘢痕内炎性细胞显著减少,成纤维细胞呈梭形,细长,呈疏松状有序排列;双丹脂质体凝胶剂组的成纤维细胞密度明显降低,与模型组比较差异有统计学意义(P<0.05);治疗后,兔血清IL-1β、TGF-β1含量较治疗前均有显著变化,与模型组及治疗前比较,差异有统计学意义(P<0.05).结论 双丹脂质体凝胶有良好的抑瘢效果,可能通过调节IL-1β和TGF-β1的表达来发挥抑制瘢痕增生的作用.

Abstract:
Objective To study the effect and mechanism of Shuangdan liposome gels on the inhibition of hypertrophic scar in rabbit ears. Methods The model of hypertrophic scar in rabbit ears was established and treated with drugs from 21 to 42 d. The level of serum IL-1βand TGF-β1 was determined. The pathological changes of the scar tissues were observed by HE staining. The index of hypertrophic scar and fibroblast density were measured. Results Compared with the model group, the infiltration of inflammatory cells was reduced and the fibroblast density was decreased with loose and orderly arrangement in Danshen liposome gel group ( P<0. 05 ) . Meanwhile, the levels of serum IL-1β and TGF-β1 were significantly changed in rabbits after treatment with Shuangdan liposome gels ( P<0. 05 ) . Conclusion Shuangdan liposome gels display an inhibitory effect on hypertrophic scar in rabbit ears, which may be related to regulation of IL-1βand TGF-β1 expression.
