经体外预处理人胎儿骨髓间充质干细胞的潜在自身成瘤性

摘    要:
目的评价经体外预处理、启动免疫保护功能的人胎儿骨髓间充质干细胞(hfBMSCs)的潜在自身成瘤性。方法采用流式细胞术检测细胞表型及细胞周期,染色体核型分析检测遗传稳定性,裸鼠皮下接种检测体内成瘤性。结果体外预处理的hfBMSCs呈正常间充质干细胞表型,可通过阻滞细胞周期于G0-G1期抑制hfBMSCs生长增殖(P〈0.05),对染色体核型无影响,不具备体内成瘤性。结论经体外预处理的人胎儿骨髓间充质干细胞不具有自身成瘤性。

Abstract:
Objective To evaluate the self-oncogenicity of human fetal bone marrow derived mesenchymal stem cells(hfBMSCs)that has been pretreated and whose immunoprotective function has been primed in vitro.Methods The cell phenotype and cell cycle were detected by FCM,the genetic stability by chromosome karyotype analysis,and the tumorigenesis in corpora by subcutaneous inoculation of pretreated hfBMSCs in athymic mice.Results The prtreated hfBMSCs displayed normal MSCs phenotype,the proliferation index of pretreated hfBMSCs was depressed and the cell cycle was blocked at G0-G1 stage(P0.05).The result of chromosome karyotype analysis revealed that this hfBMSCs had normal chromosome caryogram.No oncogenicity was observed in pretreated hfBMSCs in vivo.Conclusion The hfBMSCs has no self-oncogenicity after pretreatment in vitro.
