酸枣仁汤对小鼠试验性急性肝衰竭的影响

摘    要:目的:研究酸枣仁汤对小鼠试验性急性肝衰竭的影响。方法:雄性昆明小鼠,给予D-半乳糖胺(D-Gal-N)和脂多糖(LPS)同时腹腔注射,制备小鼠急性肝衰竭模型;并于造模前2 h,治疗组分别给予相应药物灌胃,正常、模型组给予蒸馏水灌胃。比较各组小鼠的24 h存活率,血清转氨酶水平,肝脏组织病理学改变,血清中肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)的水平,肝脏组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、谷胱甘肽还原酶(GR)、一氧化氮(NO)、一氧化氮合酶[NOS包括总一氧化氮(T-NOS)和诱生型一氧化氮(iNOS)]的水平。结果:D-Gal-N/LPS可以导致小鼠急性肝衰竭;酸枣仁汤可以提高小鼠存活率,减轻肝脏病变程度,降低血清转氨酶活性及TNF-α,IL-1β的浓度,增加肝脏组织中SOD,GR的活性,降低NOS的活性及MDA,NO的浓度。结论:酸枣仁汤可减轻D-Gal-N/LPS所致的急性肝衰竭,提高小鼠的存活率,其作用机制可能与它影响睡眠从而影响炎性细胞因子(如TNF-α,IL-1)的释放和机体氧化能力的改变有关。

Abstract:OBJECTIVE: To investigate the effect of Suanzao nacute hepatic failure in mice. METHOD: Acute liver failure was induced in male Kunming strain mice by enterocoelia injecting the animals with D-Gal-N and LPS. The mice in treatment groups were given corresponding drug 2 h before administration of D-Ga1-N and LPS, and the mice in control group were given the same dose of distilled water. The 24 h survival rate, serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) levels were compared. Serum the levels of TNF-alpha and IL-1 and the levels of SOD, MDA, GR, GSH, NO and NOS in the liver were determined. RESULT: Treatment with suanzaoren decoction could increase the survival rate and improve the liver histological feather. Suanzaoren decoction inhibited the serum the levels of ALT, AST, TNF-alpha and IL-1, and reduced the levels of MDA, NO and NOS and increased the levels of GR and SOD in the liver. CONCLUSION: Treatment with Suanzaoren decoction can suppress the D-Gal-N/LPS-induced acute hepatic failure. It may be the mechanism that Suanzaoren decocotion regulate the production of inflammatory cytokines and free radicals.
