黑色素肿瘤鼠红细胞变形能力的研究

摘    要:目的:研究患有黑色素肿瘤的小鼠在不同阶段红细胞的变形指数,松弛指数和取向指数的变化,以此研究黑色素肿瘤的发展规律。方法:人工接种黑色素瘤细胞在小鼠腿上制造实验模型,用激光衍射仪(LBY-XB2)测量长有黑色素肿瘤的小鼠在不同发展阶段红细胞的变形指数,取向指数和松弛指数,结合用原子力显微镜观察正常鼠和肿瘤鼠红细胞的形态特征加以证明。结果:实验发现,接种黑色素瘤细胞的肿瘤鼠,红细胞的变形指数是降低的,变化是显著的(P≤0.05);松弛过程中的时间常数和松弛半时间是明显增加的(P0.01),而取向指数是明显地降低的(P0.01);红细胞表面出现颗粒样物质,似有空洞发生,且细胞重叠难以分散。结论:患有黑色素肿瘤小鼠的红细胞表面是奇变的,变形能力是低的,微循环是差的。

Abstract:Objective:In this work,erythrocyte deformation index(DI),orientation index(OI) and relaxation index(RI) at differ-ent stages of tumor development in mice are investigated.Methods:The melanoma cells were innoculated on the mice's leg,for experimental model.Erythrocyte deformation index,orientation index and the relaxation index were measured by LBY-BX2 ektacytometryl on melanoma tumors at different stages of development,and proved through testing normal mice and tumor mice morphology of red blood cells by atomic force microscopy.Results:It was found that melanoma cells were innoc-ulated in mice,red blood cell deformability was reduced,that of which was significant(P ≤ 0.05);relaxation process time constant and relaxation half-time were significantly increased(P〈 0.01),while the orientation index was significantly lower(P 〈0.01);granular matters appeared on the erythrocyte surfaces,and were empty,the cell overlap was difficult to disperse.Con-clusions:The surface of red blood cells in mice with melanoma was aberrance,red cell deformability was low,microcircula-tion was poor.
